چکیده

Background and Objectives: The DNA probe assay is an alternative to culture for the detection of Neisseria gonorrhoeae which does not depend on the viability of the organism. There have been few published studies comparing the two methodologies. The majority of these studies have shown the probe assay to be comparable to culture. The goal of this study was to compare the performance of the DNA probe assay with culture in a high prevalence setting with nearly optimal culture transport conditions. Methods: Genital specimens for culture and for DNA probe assay were collected at the time of the routine visit for STD clinic patients. Results of the two methods were compared. Discrepant results were further analysed using the results of direct patient Gram stains. Results: Satisfactory matched specimens were available for 999 patients. The overall sensitivity and specificity of the DNA probe as compared with culture was 90 and 96%. Resolved values achieved by factoring in the results of direct patient Gram stains did not significantly improve the performance of the probe. Conclusion: Culture remains the test of choice for the detection of gonorrhoea in a high prevalence setting where culture transport conditions are adequate. (Genitourin Med 1996;72:108-1 10)

Incidence of Neisseria gonorrhoeae isolates negative by Syva direct fluorescent-antibody test but positive by Gen-Probe accuprobe test in a sexually transmitted disease clinic population.

To determine the accuracy of the Syva (Palo Alto, Calif.) direct fluorescent-antibody (DFA) test in comparison with the Gen-Probe (San Diego, Calif.) Accuprobe culture confirmation test, we tested 395 isolates of Neisseria gonorrhoeae from cultures obtained from patients attending a sexually transmitted disease clinic from 1 July 1991 through 30 June 1992. All isolates were tested for DFA react...

Evaluation of culture and the Gen-Probe PACE 2 assay for detection of Neisseria gonorrhoeae and Chlamydia trachomatis in endocervical specimens transported to a state health laboratory.

The Gen-Probe PACE 2 assay (Gen-Probe Inc., San Diego, Calif.) was compared with culture for the detection of Neisseria gonorrhoeae in endocervical specimens that were mailed to the laboratory. During mail transport, the specimens were exposed to extremes of hot and cold weather for several days before arriving in the laboratory. Specimens on culture plates deteriorated during transport, as evi...

Papers Evaluation of an in-house polymerase chain reaction for detection of Neisseria gonorrhoeae in urogenital samples

Aim—To develop and evaluate a one day in-house polymerase chain reaction (PCR) assay for the detection of Neisseria gonorrhoeae DNA in urogenital samples. Methods—429 urogenital specimens were tested for the presence of N gonorrhoeae by in-house PCR and by Gen-Probe. The PCR assay amplifies target sequences within the N gonorrhoeae cppB gene on the 4.2 kb cryptic plasmid, after which amplicons ...

Use of gen-probe probe competition assay as a supplement to probes for direct detection of Chlamydia trachomatis and Neisseria gonorrhoeae in urogenital specimens.

The potential for development of a cost-effective protocol for selective use of the Gen-Probe probe competition assay (PCA) in conjunction with PACE 2 for direct detection of Chlamydia trachomatis and Neisseria gonorrhoeae in urogenital specimens that would not compromise patient care was investigated. To accomplish this, PCA data from testing performed over 12 months were retrospectively revie...

Screening for Neisseria gonorrhoeae using the BD ProbeTec nucleic acid amplification test in a low prevalence genitourinary medicine clinic.

This study evaluated a population with a relatively low prevalence of gonococcal (GC) infection attending a genitourinary (GU) medicine clinic. Nucleic acid amplification testing (NAAT) screening was compared with culture. A total of 2004 male urine, 1699 female endocervical and 424 self-taken vulvovaginal swabs were screened using the BD ProbeTec GC NAAT alongside Neisserial culture. ProbeTec-...
